چکیده

In order to estimate whether mastoparan 7 specifically activates plant heterotrimeric G proteins, comparisons were made between the responses of a rice normal cultivar and a rice dwarf mutant, Daikoku dl, which has a mutation in the gene of the a subunit of a heterotrimeric G protein, to mastoparan 7. First, we found that mastoparan 7 induced the activity of the a amylase via the heterotrimeric G protein in rice embryo Iess half seeds. This observation indicated that mastoparan 7 activated the G proteins in plant cells, as does it in mammalian cells. Second, mastoparan 7 seemed to cause damages in rice cells in a G proteinindependent manner under some experimental conditions. We propose that mastoparan 7 should carefully be used for study on the plant G protein mediated

Exocytosis in chromaffin cells. Possible involvement of the heterotrimeric GTP-binding protein G(o).

The use of non-hydrolyzable analogues of GTP in permeabilized secretory cells suggests that guanine nucleotide-binding regulatory proteins (G proteins) may be involved in regulated exocytosis. Because GTP analogues are known to modulate both monomeric low molecular mass G proteins and heterotrimeric G proteins, we have examined the effect of mastoparan, an activator of heterotrimeric G proteins...

Mastoparan, a G protein agonist peptide, differentially modulates TLR4- and TLR2-mediated signaling in human endothelial cells and murine macrophages.

Previous studies have implicated a role for heterotrimeric G protein-coupled signaling in B cells, monocytes, and macrophages stimulated with LPS and have shown that G proteins coimmunoprecipitate with membrane-bound CD14. In this study, we have extended these observations in human dermal microvessel endothelial cells (HMEC) that lack membrane-bound CD14 and in murine macrophages to define furt...

Exocytosis in chromaffin cells: evidence for a MgATP-independent step that requires a pertussis toxin-sensitive GTP-binding protein.

We have previously described that mastoparan, an amphiphilic tetradecapeptide that activates heterotrimeric G-proteins, inhibits Ca(2+)-induced MgATP-dependent secretion from streptolysin-O-permeabilized chromaffin cells [Vitale, Mukai, Rouot, Thiersé, Aunis and Bader (1993) J. Biol. Chem. 268, 14715-14723]. Our observations suggest the involvement of an inhibitory G(o)-protein, possibly locate...

Mastoparan induces oscillations of cytosolic Ca2+ in rat pancreatic acinar cells.

Microfluorimetry of fura-2 was used to monitor [Ca2+]i in single cells stimulated with the G-protein activating agent mastoparan. Mastoparan induced the generation of [Ca2+]i oscillations, which in contrast to oscillations induced by low concentrations of CCK were acutely dependent on the presence of extracellular Ca2+. Oscillations were inhibited by phorbol ester. Sodium fluoride, a known acti...

NO inhibits signal transduction pathway for ATP release from erythrocytes via its action on heterotrimeric G protein Gi.

The release of ATP from erythrocytes involves a signal transduction pathway of which cystic fibrosis transmembrane conductance regulator, PKA, adenylyl cyclase, and the heterotrimeric G proteins G(s) and G(i) are components. In the pulmonary circulation, ATP released from the erythrocyte stimulates nitric oxide (NO) synthesis, thereby regulating vascular resistance. We reported that NO liberate...
